In addition, we employ the same parental CHO cell line and NS0 cells for challenging protein production in our stable expression services. This ensures a seamless scale-up process and consistent yields for your large-scale protein production. Having produced over 3,500 non-antibody proteins using these mammalian cell lines, we are a trusted partner for a wide range of research and biologics drug development innovations.

Transient Expression and Purification Services (HEK293 & CHO)

 

Utilizing our high-titer expression systems, we excel in high-throughput (HTP) transient expression and purification, employing both HEK293 and CHO cells. This allows for the rapid screening of a wide variety of molecules. Our extensive expertise encompasses the production of a diverse range of proteins, including cytokines, MHCs, TCRs, enzymes, transcription factors, as well as the extracellular domains (ECD) of cell surface receptors and membrane proteins.

Case Study: A Solution to the Challenges in Protein Engineering: Our HTP Transient Expression Services

 

In the field of protein engineering, there is often a need to screen numerous protein constructs in order to identify those that demonstrate high expression levels without any aggregates or degradation. Cost is typically a crucial factor in such projects. We offer an ideal solution to tackle these challenges through our HTP transient expression services.

Figure A: 24 constructs were produced by cloning 12 different proteins with either a His tag or Fc tag. Subsequently, these constructs were expressed in HEK293 cells utilizing 24-deep well plates. All constructs were expressed and purified in a single batch, demonstrating a rapid and efficient method for screening a large number of constructs to evaluate their expression levels.

Stable expression and purification play a pivotal role in projects that require sustained, dependable protein yields over time. Our platform harnesses both CHO and NS0 cell lines to fulfill these requirements. One significant advantage of utilizing our CHO cells lies in their lineage, originating from the same parental cell line as our transient CHO cells. This common origin facilitates smooth scale-up procedures. Moreover, this continuity extends seamlessly to CMC activities, establishing an integrated pathway from research to production. In cases where challenges arise with CHO cells, our NS0 line offers an alternative option, demonstrating robust performance across a wide spectrum of proteins.

Case Study: Proven Consistency & Reproducibility: WuXian™ Express Delivers Across All Scales

Figure A: These data illustrate remarkable consistency and reproducibility across various batches of protein production, ranging from 200 L to 2,000 L. Utilizing our CHO stable platform, WuXian™ Express, the production of the target protein in 3 distinct batches (one at 200 L and two at 2,000 L) not only showcased high consistency during the transition from medium to large scale, but also demonstrated high batch-to-batch reproducibility, as confirmed by SEC-HPLC, cIEF, SDS_CALIPER, and Glycosylation analysis.

Figure B: These data demonstrate a significant improvement in the expression of a cysteine-rich protein when utilizing NS0 cells. A 39 kDa Fc-fusion protein containing 10 cysteines exhibited aggregation and disulfide bond scrambling when expressed in CHO cells. Switching to NS0 cells resulted in a notable increase in both yield and quality. The target protein reached a purity level of 94.3% with correctly formed disulfide bonds.
