Abstract:

Targeted integration has recently emerged as a powerful approach for CHO cell line development. By enabling the precise insertion of transgenes into defined genomic loci, targeted integration generates recombinant cell lines with predictable copy numbers and integration sites, resulting in consistent productivity and long-term stability.

 

Historically, however, relatively low productivity has limited the broader adoption of targeted integration for clinical and commercial manufacturing. This limitation is largely due to the scarcity of genomic sites capable of supporting high levels of expression.

 

WuXi Biologics recently launched the WuXia TrueSite technology platform, a state-of-the-art solution for targeted integration in cell line development. The platform supports excellent product quality across a wide range of protein modalities and consistently delivers antibody titers exceeding 6 g/L from stable pools and 8 g/L from clones under fed-batch processes.

 

WuXia TrueSite also demonstrates outstanding cell line stability and strong consistency between pool- and clone-derived materials. This performance enables the production of non-GMP materials before completion of final clone stability studies, helping streamline CMC timelines to 10 months or less and accelerating the advancement of first- or best-in-class molecules into clinical development.
