Case Study 1: TCR Affinity Maturation with Rapid koff Ranking for High-Affinity, High-Specificity TCR Discovery

This case study highlights a streamlined TCR optimization workflow combining CDR engineering, high-specificity screening, and rapid SPR koff ranking to generate high-affinity, high-specificity TCRs. The optimized molecules achieved ~10,000-fold affinity improvement with strong selectivity for target over non-target, and were further used for high-titer TCR-TCE production.

Case Study 2: TCR Affinity Maturation Improves Target Binding While Preserving Non-Target Selectivity+

In this case study, the SPR data showed multiple engineered TCR variants achieving up to 14,347-fold improvement in TCR affinity for the target antigen, while maintaining no or weak binding to the non-target antigen, supporting TCR optimization with high selectivity for downstream TCR-based therapeutics development.
