WuXia RidGS System: A Rapid, High-Yield Antibiotic-free System for Stable and Scalable Biopharmaceutical Production

The GS (Glutamine Synthetase) Cell Line Development System enables efficient generation of high-yield mammalian cell lines for biotherapeutic production. By linking target gene expression to the GS gene, only cells successfully integrating the transgene can survive in glutamine-deficient media. Using ZFN technology, we achieved biallelic GS knockout in CHO K1 cells and established the WuXia RidGS™ platform with verified stability, growth performance, and product quality.

 

The WuXia RidGS™ platform shortens development to as little as 2.5 months from transfection to clone selection, achieving productivity of 7–8 g/L, and up to 10 g/L under conventional TFB conditions. It offers excellent stability (>90% top clones stable), desirable Mannose-5 levels, and strong scalability. When integrated with the WuXi Up process, expression can exceed 100 g/L, providing a robust and efficient solution for consistent, high-quality biopharmaceutical production.
